We use diverse approaches to understand the specific mechanisms cells use to successfully fold proteins into their functional, three-dimensional structures. This research is important because many human diseases occur when proteins fail to fold correctly. Our particular focus is on the earliest steps of folding, including while protein synthesis is still underway or while proteins are being translocated from one end to the other into another cellular compartment. These conditions enable protein folding to start from one end to another, which we have shown can significantly influence folding outcomes versus refolding proteins in the test tube.
